Title: THE INFLUENCE OF PLATELET-DERIVED GROWTH FACTOR AND FIBROBLAST GROWTH FACTOR 2 ON OLIGODENDROCYTE DEVELOPMENT AND REMYELINATION Author: Joshua C. Murtie Ph.D. (2004) Thesis Directed By: Regina C. Armstrong Ph.D. Professor, Department of Anatomy Physiology, and Genetics ABSTRACT: Multiple sclerosis (MS) is a demyelinating disease of the central nervous Multiple sclerosis (MS) is a demyelinating disease of the central nervous system (CNS) characterized by repeated episodes of autoimmune-mediated demyelination. Symptoms of the disease range from loss of vision to paralysis with each episode resulting in a decreased remyelination response. If remyelination does not occur, bare axons will not be able to function properly either by inefficient saltatory conduction or by degeneration resulting from a lack of myelin. This study examines the effects of growth factors on recovery from demyelination. Specifically, what roles do plateletderived growth factor (PDGF) and fibroblast growth factor 2 (FGF2) play during remyelination of the central nervous system? This question will be addressed using the cuprizone model of demyelination with significant remyelination. The remyelination response in this model will be examined in FGF2 knockout mice as well as PDGF alpha receptor (PDGFαR) heterozygous mice. This study examines the elimination of FGF2 signaling and the reduction of PDGF signaling in an animal model of demyelination with significant remyelination. The current results demonstrate that the predominant role for FGF2 during development and remyelination is that of an inhibitor

Bone Marrow Stromal Cell Transdifferentiation into Oligodendrocyte-Like Cells Using Triiodothyronine as a Inducer with Expression of Platelet-Derived Growth Factor α as a Maturity Marker

Background: The present study investigated the functional maturity of oligodendrocyte derived from rat bone marrow stromal cells (BMSC). Methods: The BMSC were isolated from female Sprague-Dawley rats and evaluated for different markers, such as fibronectin, CD106, CD90, Oct-4 and CD45. Transdifferentiation of OLC from BMSC was obtained by exposing the BMSC to DMSO and 1 µM all-trans-retinoic a...

The effect of Fibroblast Growth Factor-2(FGF-2) and retinoic acid on differentiation of mouse embryonic stem cells into neural cells

Introduction: Embryonic Stem (ES) cells as pluripotent cells derived from the inner cell mass of blastula can differentiate to neural cells in vitro and this property is valuable in studies of neurogenesis and in the generation of donor cells for transplantation. In this regard, the propose of this research, was the study of the role of two important factors in the development of neural syst...

In situ expression of fibroblast growth factor receptors by oligodendrocyte progenitors and oligodendrocytes in adult mouse central nervous system.

Basic fibroblast growth factor (bFGF) induces proliferation and alters differentiation of cultured oligodendrocyte lineage cells. In situ, bFGF is present in normal adult central nervous system (CNS) and upregulated during an early stage of various pathological conditions. We examined the expression of receptors for bFGF (FGFRs) by oligodendrocyte progenitors and oligodendrocytes in situ in nor...

بررسی تاثیر پروژسترون در بیان ژن‌های P0, S100, Krox20 در سلول‌های بنیادی برگرفته از بافت چربی در شرایط آزمایشگاهی

Background: Adipose-derived stem cells (ADSCs) have noticeable self-renewal ability and can differentiate into several cell lines such as adipocytes, osteoblasts, chondrocytes, and myocytes. Progesterone plays a significant role in the myelination of peripheral nerves. Regarding the role of progesterone on the myelination of peripheral nervous system, we evaluated its effect...

Pre-oligodendrocytes from adult human CNS.

CNS remyelination and functional recovery often occur after experimental demyelination in adult rodents. This has been attributed to the ability of mature oligodendrocytes and/or their precursor cells to divide and regenerate in response to signals in demyelinating lesions. To determine whether oligodendrocyte precursor cells exist in the adult human CNS, we have cultured white matter from pati...
